RNA polymerases:

·  Detection and separation of the three RNA polymerases (F10.2, F10.5).

·  Roles of the three RNA polymerases ­ which ones transcribe which RNAs (F10.7, T10.1).

·  RNA polymerase structure (T10.2); epitope tagging (F10.9); requirement and stoichiometry of Pol II subunits (F10.10; T10.3); RNA pol II subunit function for DNA binding (F10.12), transcription initiation at a promoter (F10.13, F10.15), elongation with phosphorylated rbpIIa (F10.17) and alpha amanatin resistance.

 

Promoters:

·  Class II core promoters (F10.25).

a)    TATA box characteristics; TATA-less promoters and where they are found; TATA box function (F10.26, F10.28, F10.29).

b)    Upstream element characteristics; GC boxes and their function (T10.4); CAAT boxes and their function.

c)    Initiator sequence and function.

 

·  Class I core promoters.

a)    Core element and upstream control element (F10.32); spacing among elements (F10.33).

 

·  Class III core promoters.

a)    Internal promoters and where they are found (F10.34, F10.35); A, B and C boxes and intermediate element (F10.36).

b)    Polymerase II-like promoters and where they are found (F10.37).
